We couple a laser beam with a wavelength of 980 nm into the fiber optic probe and manipulate the fiber to achieve the capture and transport of particles and cells with three different refractive indices in liquid： silicon dioxide （SiO 2 ）， polystyrene （PS）， and yeast cells， and thus achieve the classification of different particles in the range of 1-10 μm. The different capture capabilities of this parabolic fiber optic probe for the three particles were simulated， and the obtained theoretical and experimental results were in agreement. The use of this method to classify particles simplifies the experimental setup and has a wide range of potential applications in the development of label-free hybrid fiber optic sensors， infectious disease detection， and cell classification. 光纤光镊 折射率 细胞筛选 微粒分类 fiber optic tweezers refractive index cell screening particle classification 1 引 言 1 微粒、活细胞和大分子的无触点和无创分选是多学科研究的一个主要目标，特别是在生物医学和化学分析方面 ［ 1 1 ］ 。近年来，光镊技术已经在微米尺度量级粒子的操纵控制、粒子间的相互作用等方面发挥了重要作用 ［ 2 2 - 5 5 2-5 ］ ，这一技术也受到了越来越多的关注 ［ 6 6 - 7 7 6-7 ］ 。光学镊子于1986年由Arthur Ashkin首次提出利用光压操纵微小粒子的概念 ［ 8 8 ］ 。光镊对微粒的操控是非接触的，不会给操作对象造成机械损伤。这使得光镊非常适合应用在生物学研究中，尤其是单细胞、单分子研究领域 ［ 9 9 - 10 10 9-10 ］ 。传统的光镊装置都是基于笨重的光学器件 ［ 11 11 - 13 13 11-13 ］ ，使用时存在一些限制，比如实验中会用到的装置较复杂 ［ 14 14 - 15 15 14-15 ］ ，需要昂贵的微加工成本等 ［ 16 16 - 17 17 16-17 ］ 。现代生物学的发展迫切需要可以简单、高效地操纵微、纳米大小的粒子和细胞的工具。 光纤光镊因其结构紧凑、便于集成、操作灵活、适用范围广等特点，成为广泛应用于光学捕获和光学操纵的工具。基于光纤的光镊系统，其灵活性和微型化等特点，为医学和生物学研究微粒分类提供了更好的解决方案 ［ 18 18 ］ ，在生物医学、化学等学科进行微粒分类时能够快速、准确地完成分类 ［ 16 16 ］ ，在相关领域已取得重大进展 ［ 19 19 - 20 20 19-20 ］ 。2011年，Wang， X.， Chen， S.等人整合了光学镊子和微流控芯片技术，用于处理具有高精确度的小细胞群分选。微流控技术的层流特性使目标细胞集中在所需区域进行细胞分离。同时为了识别目标细胞，开发了一种图像处理方法，具有多种特征的识别能力。目标细胞可以通过光学镊子以非侵入性的方式精确移动到目标位置。这种分类功能的独特优势在于其在小细胞群分类中的高精度 ［ 21 21 ］ 。2012年，李宝军课题组利用光纤在流体中分离两种不同大小的颗粒。当波长为1 550 nm激光入射光纤后水中的粒子在光引力的作用下被大量捕获并聚集在光纤周围。通过利用流体流态和作用在微粒上的梯度力，将组装好的微粒按照直径大小分类并放置在不同区域。该分类方法的关键因素是光纤中泄漏的光的强度分布和能量分布的不对称性 ［ 22 22 ］ 。2018年，Paiva等人利用光纤尖端顶部聚合物形成的聚合透镜，通过对捕获粒子后向散射信号的45个特征集的探索性分析，创建了一个新的单一特征，能够区分合成粒子和活酵母细胞 ［ 23 23 ］ 。2018年，Rodrigues等人提出了一种基于单模光纤顶部的聚合物微透镜的光纤镊子结构。这种制造方法能够产生利用率更高的光纤针尖，对目标产生更强的捕获效应，捕获能力可提高2~10倍。 利用后向散射信号这些透镜拥有了区分不同粒子的能力 ［ 24 24 ］ 。2021年，Ai等人利用光镊在空气中捕获单个微粒，并使用拉曼光谱来检测和鉴定空气中的微粒类型。利用嵌入式成像系统实时显示捕获微粒的形状和尺寸 ［ 4 4 ］ 。这些方法虽然可以完成对微粒的分类，但在实际操作过程中存在着实验系统搭建难度大、分类所需设备复杂、要求微粒直径相同、对光纤尖端尺寸要求高或采集的信号加工处理困难等问题。因此，简化光学分类技术和扩大其适用性是在溶液中捕获微粒并进行分类领域中亟待解决的问题。 本文提出的利用热熔法制作的抛物线型单光纤探针在液体中对不同折射率的微粒具有不同的捕获能力，实现了对二氧化硅、聚苯乙烯和酵母菌细胞的分类，在对SiO 2 和酵母菌细胞进行分选时，通过形状的差异对两种微粒进行区分。由于微粒或微生物细胞的化学组成往往会产生不同的折射率，因此，不同折射率的微粒光学分选对于化学和生物医学分析应用范围更大。经过实验验证，该结构光纤可广泛应用于微粒和细胞筛选、分类领域。实验所需光学设备不复杂、易操作，并可准确的实现对微粒和生物细胞的分类。研制一种尖端可以捕获、操控和对微粒进行分类的光纤将是对生物医学细胞及微粒研究的一大进步。 2 基本原理及实验仿真 1 2.1　理论计算方法 2 光学力一般由辐射压力产生，光照向微粒，发生折射引起动量的转移。光学力包括梯度力和散射力。梯度力能够捕获微粒，散射力能够驱赶微粒远离光纤顶端。梯度力又包括横向梯度力和轴向梯度力，横向梯度力的微粒拉向光轴，轴向梯度力则把微粒拉到光场最强处。通过横向梯度力和轴向梯度力的共同作用，微粒能够被操控、捕获 ［ 26 26 ］ 。 单独一束激光的光学势阱捕获取决于入射光波长、捕获对象尺寸、对象的折射率和周围介质折射率之间的关系。对于粒子尺寸远大于激光波长时，光学梯度力可以根据几何光学模型计算，也是分析折射率与动量守恒之间的关系。对于粒子尺寸远小于激光波长时，粒子被视为不均匀电磁场中的电偶极子，根据瑞利散射机制进行分析 ［ 27 27 - 28 28 27-28 ］ 。当粒子尺寸在0.1~10 λ 2.2　仿真结果 2 基于动量守恒原理计算了不同折射率微粒的光捕获力。利用商业有限元仿真软件中的“电磁波频域”界面，建立二维模型，并分别计算了酵母菌细胞、SiO 2 和PS微粒的俘获力。在二维模拟模型中，背景溶液的折射率为1.33，所用的SiO 2 ，PS形状为圆形，折射率分别为1.46和1.59，微粒的大小分别为5 μm和3 μm，酵母菌细胞为椭圆形，折射率为1.42，长轴为5 μm，短轴为3 μm。入射光源波长为980 nm，入射激光功率为5 mw。如 图1 图1 所示，对微粒及酵母菌细胞的光场进行仿真，对其受力情况进行了分析。图 1 1 （a）~ 1 1 （c）是抛物线型光纤对SiO 2 微粒捕获的仿真示意图。由 图1 图1 （b）可知，轴向力大于零，这意味着SiO 2 微粒被推动沿着光束的传播方向移动，远离光纤尖端。轴向力小于零，这意味着SiO 2 微粒被拉动沿着光束传播的反方向移动，靠近光纤尖端。因此，SiO 2 微粒在轴向上存在一个稳定的捕获点。然后，计算了该捕获点施加在SiO 2 微粒上的横向力（见 图1 图1 （c））。由于光场沿 x 轴对称，可以在 x 轴上捕获粒子，因此粒子受到的横向捕获力是中心对称的，如 图1 图1 （c）所示。当施加在SiO 2 微粒上的轴向捕获力和横向捕获力为零时，粒子保持平衡。由于PS微粒的受到的轴向力大于零，光阱不能捕获微粒，在轴向PS微粒会被推走。在横向上PS微粒受到横向的作用向光轴方向运动，如图 1 1 （d）~ 1 1 （f）。从图 1 1 （g）~ 1 1 （i）可以看出，同样，酵母菌细胞的轴向力与SiO 2 轴向力的受力情况类似，在光纤出射光场的焦点上可以被稳定捕获（见 图1 图1 （h））。从 图1 图1 （i）中可以看出，微粒由于横向力的作用被吸附到光阱中。在分类的过程中，从形状及特性上可以分辨SiO 2 微粒和酵母菌细胞。因此，从仿真受力中可以看出，单光纤光镊可以捕获SiO 2 微粒和酵母菌细胞，并推走PS微粒。 为了进一步验证理论的正确性，根据上文仿真结果，使用仿真软件对直径不同的微粒进行了仿真，并计算不同尺寸微粒受到的光阱力。图 2 2 （a）~ 2 2 （c）显示了直径不同的SiO 2 ，PS微粒和酵母菌细胞的轴向光阱力；由图 2 2 （a）~ 2 2 （c）可知，SiO 2 和酵母菌在轴向上基本上都有稳定的捕获点。但是，由于PS受到的轴向力都大于零，所以PS会沿着光束传播的方向远离光纤尖端。图 2 2 （d）~ 2 2 （f）计算了直径不同的SiO 2 ，PS微粒和酵母菌细胞的横向光阱力。由图 2 2 （d）~ 2 2 （f）可知，不同尺寸的SiO 2 ，PS和酵母菌微粒受的横向力在 x 轴上达到平衡。也就是说，只要微粒偏离光轴，就会在横向力的作用下被拉回光轴。因此，该单光纤光镊可以实现对不同直径大小的微粒捕获，传输和分类。 2.3　光纤的制备 2 实验中用到的光纤探头采用热熔法拉伸制作，使用单模光纤用火焰加热技术制造（接口类型：FC/PC，纤芯直径：9 μm，包层直径：125 μm）。第一步，使用光纤剥离器将光纤的缓冲层和聚合物夹层剥离，得到直径125 μm，长度为3 cm的裸纤。为了提高纤维的稳定性，采用不锈钢毛细管包覆。第二步，将裸纤放置在酒精灯火焰下加热至光纤的熔点，初始速度大约为5 mm/s，纤维在1 cm的长度范围内逐渐变细，直径由125 μm减小到10 μm。然后加快光纤的拉伸速度至20 mm/s，在尖端形成一个锥形区域，之后光纤被折断，尖端变细。熔化纤维的表面张力在末端形成了锥形尖端。再用一块酒精棉仔细擦拭锥形纤维的抛物线型尖端，最终得到的光纤尖端如 图3 图3 （a）所示。第三步，利用上述步骤得到的抛物线型光纤，在配置好的混合溶液中捕获微粒。该抛物线型光纤探针的制作方法简单，成本较低。 3 分析与讨论 1 3.1　实验装置 2 如 图3 图3 所示，搭建实验装置进行实验，验证理论的可行性。采用波长为980 nm的激光器（YA605，nbyebo）作为光源，输出功率为5 mW。光源通过光纤（SW630-125-13/250）传输到光纤尖端，光纤尾部FC/PC接头连接到激光器的输出端，实现对微粒和生物细胞的操控和运输。为了减少实验中由于环境波动而产生的振荡，光纤探针由毛细管包裹被固定在三维操作台上。将载玻片安装在 xyz 手动平移台上确保定位的精确性和稳定性。这是利用带有CCD相机的计算机搭建的可视化分类过程和捕获图像过程的实验系统。为实现单光纤光镊精确操控PS，SiO 2 微粒和酵母菌细胞进行分类，将微球悬浮液滴在安装在三维微平台的载玻片上，同时将光纤探针浸入微球悬浮液中。 所用的混合溶液由微粒和生物细胞在蒸馏水稀释而成。悬浊液放置在盖玻片与载玻片之间，中间留有2~3 mm的距离形成腔室，以减弱微粒在溶液中的布朗运动，可以1~2 h内工作在一个稳定的环境下。腔室固定在三维精密操作台上，以便对光纤尖端进行校准。混合溶液中的微粒及生物细胞通过单光纤光镊进行操控，完成传输和分类。这个过程由CCD相机进行成像。混合溶液的微粒直径大小为1~10 μm，且三种微粒的折射率大小不同。 3.2　实验结果 2 搭建实验所需要的实验装置，如 图4 图4 所示，利用折射率的不同实现对微粒和细胞的分类。在实验中，使用激光波长为980 nm的光纤，光纤探针可以操作大小在1~10 μm的细胞和微粒。首先配制出直径在1~10 μm的PS微粒、SiO 2 微粒和酵母菌细胞的混合溶液。粒子为悬浮态，三种混合微粒在液体中随机分散。图像由CCD采集并在计算机上实时观察。光纤探头放入细胞与颗粒的混合溶液中，利用三维台实现光纤探头的三维移动，固定激光功率为5 mW。通过调节三维台，使用光纤在混合溶液中操控微粒的运动状态，完成对酵母菌、SiO 2 和PS三种微粒的传输与分类。 图5 图5 （a）和 图5 图5 （b）为微粒分类模型图； 图5 图5 （c）是光学显微镜下的分类结果，将波长为980 nm的激光束入射到光纤中，形成稳定的光阱。由于光纤探针对不同折射率的微粒的捕获效果不同，进而了SiO 2 ，PS微粒和酵母菌细胞的分类。由于样本的局限性，SiO 2 ，PS微粒和酵母菌细胞的粒径在1~10 μm基本上都可以捕获。在对SiO 2 和酵母菌细胞进行分选时，主要是通过形状和微粒的特性对两种微粒进行区分。图 5 5 （d）~ 5 5 （f）显示了不同折射率的微粒通过光纤尖端的分类过程。图 5 5 （d）， 5 5 （f）显示了移动三维台操控光纤靠近目标微粒。当光纤尖端靠近微粒时，由于光纤的光阱力的作用，直径为1 μm，3 μm，4 μm，6 μm，10 μm的酵母菌细胞和SiO 2 微粒就会被稳定捕获，并且停留在光纤的焦点附近。在捕获微粒以后，匀速缓慢移动位移台，溶液中的微粒就会跟随着光纤移动，从而改变微粒的位置，实现对微粒分类的目的。 图5 图5 （e）是使用抛物线型光纤推走PS微粒。由于PS微粒受到的轴向力为正，直径为1~9 μm的PS微粒会被推走。通过改变该光纤的位置及激光功率的大小，操控微粒到达放置区域。在实验过程中，通过操作三维台控制光纤位置时，要防止移动速度过快导致微粒和细胞脱离光阱力的作用而掉落。同时，固定激光功率为5 mW，防止在操控PS微粒的过程中，位移的速度过快，未能达到微粒放置的指定区域。通过理论计算和实验验证，证明了所提出的光学探针可以同时在混合溶液中完成对微粒和细胞的分类。 在实验过程中，通过调节不同的激光功率对混合溶液中微粒进行捕获和操控，获得光纤光镊的最佳捕获和操控的效果。同时为了防止激光功率过大对细胞造成损伤，因此要找到对微粒和细胞分类所需要的最合适的功率。如 图6 图6 所示，通过对实验结果进行分析，得到了不同激光功率捕获微粒的稳定性及操控微粒的速度。 如图 6 6 （a）和 6 6 （b）所示，分别使用不用的激光功率对SiO 2 微粒和酵母菌细胞进行捕获。在功率为1mW和3mW时，微粒会产生明显的震动，在捕获时微粒容易脱离光阱。当功率调节到5mW时，微粒在捕获过程中受到较大的梯度力，微粒可被稳定的捕获，并且在移动光纤时微粒较为稳定的移动。从图中可以看出功率越大，微粒和细胞所受的梯度力就越大，光阱对微粒的束缚力就越强，捕获微粒就更加稳定。但是功率越大对细胞的损伤就越严重，因此，选用功率为5 mW能够稳定捕获细胞，并且减少光损伤。如 图6 图6 （c）所示，由于PS微粒受到的散射力比较大，因此光纤尖端可以推动PS微粒到指定的区域。当激光的功率增大时，PS微粒的位移速度会增加。通过计算在功率为5 mW时，微粒的位移平均速度为1 μm/s，易于操控PS微粒到达指定位置。 由实验与仿真结果可知，本文提出抛物线型的光纤探针实现了静态液体中对不同折射率微粒的分类。对比其他细胞分选技术，使用单光纤光镊的细胞分选技术优点明显，在细胞分选过程中可以使用活体细胞，而且不需要进行液相处理。因此光镊细胞分选技术的分选环境更接近细胞生存的环境，能很好的保护细胞的活性和结构特性。基于不同折射率的微粒对微粒进行分类，可以在混合溶液中准确的捕获所需要的微粒，以便于微粒或者细胞进行后续研究。 4 结 论 1 本文提出了一种采用熔融法拉伸的抛物线型光纤探针，实现对不同折射率微粒的分类。利用熔融法得到的特殊的抛物线型光纤探针，对于浸没在溶液中不同射率的微粒具有不同的操作能力，以达到微粒分类的目的。理论上，通过仿真验证了这种抛物线型光纤探针对不同微粒具有不同的捕获能力。在实验中，通入波长为980 nm激光，实现了混合液体中三种不同折射率的微粒及生物细胞的运输与分类。在对SiO 2 和酵母菌细胞进行分选时，通过形状的差异对两种微粒进行区分。理论和实验结果都表明，当微粒折射率不同时，可以利用折射率的差异实现1~10 μm微粒的光学分选。使用本文所提出的对微粒的光学分类方法，有望应用于生物样本的分离和运输，实验室的化学或者生物医学分析等方面。在分类过程中，准确率较高，并且所需要的设备简单，造价较低。 参考文献 DHOLAKIA K ， ČIŽMÁR T .
